A 42-year-old man developed optic neuropathy, and experienced irreversible visual loss, while receiving disulfiram [dosage, route and indication not stated]. He experienced progressive bilateral visual loss 6 months after starting disulfiram. His mitochondrial DNA was sequenced to revealed the homoplasmic mutation m.11778G>A, which is associated Leber's hereditary optic neuropathy (LHON). In this patient, disulfiram was associated with increased cell death in cells containing a high mutation load.Author Comment"We identified a patient with disulfiram-induced bilateral optic neuropathy who was subsequently shown to carry a mtDNA mutation associated with LHON."
