In a prospective observational cohort study (DRKS0001 0502) reviewing cases from the German embryotox pharmacovigilance institute database identified 14 neonates· (8 male and 4 female) [ages not stated; not all sexes stated] who developed various toxicities following inutero exposure with captopril, enalapril, lisinopril, perindopril or ramipril during the first trimester [routes not stated; not all dosages and outcomes stated].
Pregnancy 1: A neonate developed potter sequence during in-utero exposure with captopril. The neonate's mother was a 17-year-old girl, who received captopril 50mg during 0-12 weeks of gestation. She also received various other concomitant medications. In view of fetal anomaly, at the 17+4 weeks of gestation, she underwent pregnancy termination due to the observed potter sequence congenital anomaly in neonate.
Pregnancy 2: A male neonate developed multi-cystic renal dysplasia and simian crease during in-utero exposure with enalapril. The neonate's mother was a 41-year-old woman, who received enalapril during 0-8 weeks of gestation. She also received metoprolol as concomitant medication. At the 37+6 weeks of gestation, she delivered a 2700g male neonate who had multi-cystic renal dysplasia (congenital cystic kidney disease right, left mild dilatation) and simian crease congenital anomalies.
Pregnancy 3: A male neonate developed congenital hydronephrosis and ureteral stenosis during in-utero exposure with ramipril. The neonate's mother was a 37-year-old woman, who received ramipril during 0-4+5 weeks of gestation. She also received various other concomitant medications. At the 33+4 weeks of gestation, she delivered a 2520g male neonate who had congenital hydro-nephrosis and ureteral stenosis.
Pregnancy 4: A female neonate developed various congenital anomalies during in-utero exposure with ramipril. The neonate's mother was a 22-year-old woman, who received ramipril 2.5mg during 0-8 weeks of gestation. She also received various other concomitant medications.
