Abstract
目的 探讨人类第9号染色体短臂(9p)上微卫星改变与肺癌发生及转移的相关性。方法 应用聚合酶链反应(PCR)结合微卫星银染分析方法检测32例非小细胞肺癌(NSCLC)组织中两个微卫星标志物IFNA和D9S171的改变。结果 32例NSCLC患者中,14例(43.8%)出现了微卫星标志物改变。其中伴有淋巴结转移的肺癌组织中微卫星改变检出率为64.7%,不伴转移的肺癌组织中微卫星改变检出率为20%,两组间比较有显著性差异(P<0.05)。16例良性肺部疾病组织均未检出微卫星改变。结论 人类第9号染色体微卫星改变在NSCLC中普遍存在,有可能成为肺癌早期诊断和转移判断的分子标志物。
Objective To investigate the relationship among microsatellite alterations on chromosome 9(9p) and the oncogenesis,metastasis of non-small cell lung cancer (NSCLC).Methods Two microsatellite markers,IFNA and D9S171,were analyzed in 32 cases of NSCLC by polymerase chain reaction-based silver-staining assay.Results Of 32 cases of NSCLC,14(43.8%) had microsatellite alterations.The alteration frequency was 64.7% in NSCLC with lymph node metastasis,and 20% in those without lymph node metastasis.There was significant difference between the two groups.None of benign lung tissues showed microsatellite alterations.Conclusion Microsatellite alteration on chromosome 9p in NSCLC appears to be a common and specific event,and may be a valuable molecular marker for determination of early diagnosis and metastasis of NSCLC.
